Data Independent Acquisition
What is DIA?
Conversely to strategies like SRM or PRM, Data Independent Acquisition, or DIA, requires no precursor information. Precursor ions are isolated in small, pre-determined isolation windows (usually 2-25m/z) and fragmented. The fragment ions of this isolation window enter the are scanned systematically across a range of m/zs (400m/z-1200m/z, for example). Because no prior knowledge of peptides or proteins is required for isolation, this technique is suited for discovery-based proteomics or profiling an obscure proteome.
